Could Sourcesafe affect IIS?

I upgraded version 2.x to 3.0. About that time, port 80 stopped receiving connections from outside of the LAN. Could sourcesafe have caused this?

Do you mean Vault, rather than SourceSafe?

Can you connect to port 80 from within the LAN?

Vault uses IIS but shouldn't change IIS connectivity. It could be that some configuration on your network has changed.
